Specifically, when yeasts are unable to remove/retrotranslocate misfolded ER membrane substrates— which i have shown to be mediated by Derlin Dfm1—E3 ligase Hrd1 can actually restore retrotranslocation! A surprising feature of this new pathway is that Hrd1 no longer employs

its partner protein nor its ubiquitin ligase activity to remove misfolded membrane substrates from the ER. This implies the Hrd1 machinery has unanticipated plasticity and breadth of function and can be completely remodeled to meet the changing stress of the ER!
