Duke et al. used galactose oxidase for site-selective oxidation of C6 primary hydroxyl on the terminal galactose residues in CPS. Immunization with the ​homogeneous glycoconjugate ​displayed an improved humoral response.

The traditional method for CPS derivatization involves periodate oxidation to generate aldehyde reactive groups for functionalization with carrier proteins. However, the method suffers from heterogeneity as well as structural changes and degradation of CPS.

Polysaccharide antigens alone only elicit short-lived IgM antibody response since they cannot be displayed by the MHCII. However, covalent conjugation of polysaccharides to protein carriers can be presented by MHCII to trigger CD4+ T cells activation.

The capsular polysaccharide (CPS) of Streptococcus pneumonia, a pathogenic bacteria with multiple serotypes, is known to be important for virulence and immune evasion. Thus, the current pneumococcal vaccine formulations use CPS as antigens.
