Using the mutant proteasomes we show that some active site selective inhibitors are partial inhibitors of the chymotrypsin site, whereas others achieve full inhibition.

We generated mutant recombinant proteasomes that show that the standard commercially available chymotrypsin substrate is also turned over by at least one of the other active sites, which makes interpretation of dose response curves challenging.
