8/10 In conclusion, our work provides an innovative framework for dissecting the crosstalk between gene regulation and inter-tissue communication in mammalian embryonic development.

7/10 Finally, we used chimeric gastruloids derived from a mix of wild-type (WT) and MSGN1 KO cells. This revealed that MSGN1 KO cells can form spinal cord in the presence of WT cells. This indicates an underlying inter-tissue communication program driven by MSGN1.

6/10 Surprisingly, MSGN1 KO not only impaired mesoderm development, as anticipated, but also negatively affected spinal cord development. This reveals an inter-tissue role of MSGN1 that was previously unknown.

5/10 We were particularly struck by MSGN1 and CDX2 as potential drivers of the early decision from neuromesodermal progenitors (NMPs) to mesoderm and spinal cord, respectively.

4/10 Next, we investigated which transcription factors (TFs) are active during gastruloid development. To do this, we defined the developmental trajectories in gastruloids and predicted TF activity based on their motif accessibility.

3/10 We discovered that the chromatin regulatory profile of gastruloids strikingly matches that of post-gastrulation embryos.

2/10 We utilized single-cell ATAC-seq to analyze developing mouse gastruloids, successfully defining their chromatin regulatory profile.

1/10 Our research aimed to answer two key questions: how do pluripotent cells make early cell fate decisions? and how does gene regulation drive these decisions?.

Have you ever wondered how your back formed? The human embryo makes a neural tube (future spinal cord) and somites (trunk muscle/bone) from ~d20. They’re formed at the same time and place, so we used human Trunk-like Structures (hTLS) to investigate their ‘co-development’… (1/7)
