One more CRISPR story we just posted on Biorxiv. Adding 5' modifications to donors stimulates HDR and donor potency. Probably does so by preventing end joining reactions that reduce available donor in cells. Please pass along.

Don't forget to optimize your RNP concentration using a co-injection marker! It is toxic, you will still get short edits with ssODNs but you won't get long edits if the conc. is too high. Worms getting long dsDNA seem to be more sensitive to RNP dose.

NEW CRISPR protocol: https://t.co/xCXKKYmtbr Please share with your friends and let me know how the hybrid donors work for you! The idea is two PCR products one with homology arms, one without, mix melt and anneal. The hybrids improve efficiency of GFP-size inserts 5-10X

The pew meeting in Tucson was fabulous! The guest speakers reminded us, our society relies on us! The talks were diverse, concise, and inspired! And although the skit was quite dumb, we had lots of fun. We're just bummed that Torsten retired. (Inspired by Anne Churchland)
