(5/6) We propose a new model in which cohesin does not need NIPBL for chromatin association but it does for loop extrusion. While cohesin-STAG1 binds at CTCF sites and becomes stabilized rapidly, cohesin-STAG2 bind elsewhere and require NIPBL in order to reach CTCF positions.

(4/6) But. can it form loops?. No, Hi-C analyses on NIPBL KD cells show loss of cohesin-mediated loops. This is not simply due to reduced cohesin on chromatin, since STAG2 KD cells display similar distribution on cohesin but long loops are formed.

(3/6) Where in the genome is this cohesin-STAG1 located? . Calibrated ChIP-seq analyses reveal that cohesin-STAG1 accumulates at the same CTCF-bound sites in NIPBL KD and control cells. Thus, cohesin-STAG1 may associate with chromatin at CTCF sites

(2/6) Using a flow cytometry assay to measure chromatin bound proteins we found that NIPBL KD decreases cohesin-STAG2 on chromatin but increases cohesin-STAG1. This suggests that cohesin-STAG1 may associate with chromatin independently of NIPBL.
