We characterized two genetic targeting systems with modified VoltageFluor dyes (synthesized by @vincentngrenier) that bind to either SpyTag or HaloTag. VoltageSpy is a little more sensitive, while VF-HaloTag gives better staining, and we demonstrate it in cultured neurons.

Earlier in my time in the Miller lab we showed that you can estimate resting membrane potential with VoltageFluor dyes and FLIM. https://t.co/75EpJ2yaDT But indiscriminate staining of all membranes makes looking at complicated cultures difficult β enter targeted voltage dyes.

This is a valid concern that comes up frequently. Two things: 1. People will look at imaging data associated with papers. We certainly require and look for such datain our lab. 2. There are solutions. See this article by Kayla Jackson:
